Abstract
The main by-product of biodiesel production, which in recent years has rocketed, is raw glycerol. Per ton of produced ethyl or methyl esters of fatty acids about 100 kg of raw glycerol was obtained. Utilization of raw glycerol is one of the most important and serious challenge in the environment protection. The aim of the study was to verify the production possibility of the cell biomass yeast Candida pelliculosa G4KB2 on pure glycerol in shake-flask cultures. In the first stage of the study, ability of growth on pure glycerol yeast Candida pelliculosa G4KB2 was checked by passaging them in a modified agar Petri dishes – YPDG (YE, peptone, dextrose, glycerol) then YPG (YE, peptone, glycerol). Cultures incubated at 30°C for 2 days. Effect of glycerol concentration on the growth rate and biomass of G4KB2 were performed in duplicate in media containing: 2, 5, 10, 15 and 20% of glycerol. Cultures were incubated at 30°C on an orbital shaker (120 rpm). The growth rate of the yeast was analyzed by measuring the optical density and biomass concentration was determined gravimetrically. The highest biomass growth (13.9 g·dm-3 at 24 h and 19.2 g·dm-3after 40 h) was obtained during culture in medium containing 10% of pure glycerol.
